Abstract

In this report an experiment is described that evaluates the use of a gene editing system suitable for the evaluation of the effect of HAT and HDAC inhibitors on the efficiency of the CRISPR-Cas9 technique. The system consists of H27 cells, which are HeLa cells that stably express eGFP gene. It is desired that the eGFP gene is knocked-out and the eYFP gene from a donor plasmid is knocked in with homology directed repair. The knock-in efficiency is measured with plate reader and FACS, respectively. It is concluded that no knock-in takes place, but knock-out does take place. The system is insufficient for the evaluation of the effect of HAT and HDAC inhibitors on the efficiency of the CRISPR-Cas9 technique. It is recommended that a different system is used.
