Abstract
Bacteria in the human gut have shown to have a relation with human health and diseases. The effects of different nutrients on these bacteria can be studied in a co-culture system with anaerobic bacteria and oxygen requiring human epithelial cells. This research project is based on this system. To be able to do faster research, faster analysis of the culture systems is required. Using image analysis, an algorithm was developed to automatically follow bacterial growth on agar plates over time. A start has been made with an algorithm for tracking oxygen penetration and pH value tracking was also of interest. The developed algorithm showed a high performance in selected test settings for determining bacterial areas. Furthermore, improved designs for culture containers and a design proposal for a robotic setup are made that are required to automatically analyze multiple culture systems in a high throughput manner. The designs require further development in future work. This research project functions as a step up for more research on these topics.
