Abstract
Hematologic malignancies occurs when hematopoiesis fails, and include lymphomas, myeloid and lymphoid leukemias. Standard treatment for these diseases is chemotherapy, but the problem with this is high relapse rates. Alternative therapy options are needed to increase survival rates and reduce the amount of relapses. An alternative treatment that has gained interest in the last couple of years is immunotherapy. A promising form of immunotherapy is the use of CAR T-cells, which are chimeric antigen receptors linked to cytotoxic T-cells. The CAR can specifically target tumor-associated antigens, and thus eliminate tumor cells via activation of the attached T-cell. Many studies and clinical trials have examined the potential therapeutic effects of CAR T-cells in lymphomas and lymphoid leukemia. In myeloid leukemia, only a few studies and clinical trials have been conducted to examine the therapeutic effects of CAR T-cells in this disease. Overall, CAR T-cells provide a very promising treatment for lymphomas and lymphoid leukemias, while they are harder to apply in myeloid leukemias, due to high heterogeneity in markers among patients. Although CAR T-cell therapy can be applied in lympomas and lymphoid leukemias, there remain serious disadvantages of current CAR T-cell therapies, such as resistance, side-effects and individual engineering. More research will expand knowledge on this, and will improve therapeutic value of CAR T-cell therapy.
