Abstract
The current computer-assisted semen analysis method used by the UMCG is not always reliable and prone to technical errors, this project focusses on validating a new manual counting method to determine sperm concentration. Therefore the exact number of counted spermatozoa per field of view was counted to determine the concentration, via a calculation method that converts a number of counted spermatozoa per field into a number of spermatozoa per ml. It was determined how many fields of view per sample have to be counted in order to reach a reliable estimation of the concentration (<25% deviation of the real concentration), distinguishing between used objective (20X/40X) and location in a counting chamber. A separate part of the project was data analysis of a database containing data about semen motility parameters and fertility outcomes (fertilized oocytes/ achieved pregnancy), in an attempt to discover a potential correlation between the parameters and fertility outcome. This data analysis was performed in R studio.
