Abstract

Alzheimer’s Disease is a progressive neurodegenerative disease, and the most commoncause of dementia. It has been shown that TNF-α plays an important role in development and progression of the disease, but the exact role is not clearly understood. Previous research has focussed mainly on deletion of TNF-α or its receptors, which leaves a need for more research regarding the effects of elevated TNF-α levels in the brain and the dynamics of the TNF-receptors in an environment of chronically elevated TNF-α. The current study focusses on evaluating the effects of elevated TNF-α levels in certain brain regions on Aβ based Alzheimer’s disease symptoms in mice. The primary outcome parameters of the study are cognitive phenotype, Aβ plaque development and neuroinflammation. From the current preliminary results it seems likely that an increase in TNF-α levels results in neuroprotective responses via the TNFR2 receptor. However, further results regarding neuroinflammatory responses and TNFR1 and TNFR2 dynamics are needed to draw a more complete conclusion.
