Abstract
In this study we successfully attempted to express three fungal type III PKSs, out of which two were active (HargPKS and AiizPKS) in S. cerevisiae. Further structural elucidation proved that the main products produced by HargPKS and AiizPKS were tri- and tetraketide pyrones. HargPKS was thereafter cloned into multigene plasmids containing the putative tailoring genes of the P. fici BGC. We evaluated the activity of the putative tailoring enzymes on products of HargPKS, products of PfPKS and other polyketide products. Additionally, the proteins produced by S. cerevisiae containing multigene plasmids were analysed using SDS-PAGE. We performed reverse transcriptase PCR to see the transcription of the genes of interest. This study serves as a basis for expressing fungal type III PKS BGCs in S. cerevisiae.
