Abstract
Tissue clearing is done to enable detecting signals from fluorescent labels deep within a specimen, permitting three-dimensional imaging of structures within these specimens. Firstly, this study explores the temporal and spatial evolution of transparency during the clearing process, focusing on the diffusion mechanisms of optical clearing agents, such as benzyl alcohol benzyl benzoate. Secondly, the quantitative influence of tissue composition on this diffusion process is investigated, which was previously largely unexplored. Thirdly, optical tissue clearing is applied on a porcine thyroid to investigate the compatibility of hydrophobic optical tissue clearing with (multi)spectral optoacoustic imaging.
