Abstract
Chronic obstructive pulmonary disease (COPD) is a progressive lung disease in which viral infections, particularly human rhinovirus (HRV), are major triggers of exacerbations. To study virus-induced exacerbations, physiologically relevant models such as airway-on-chip are needed. Before developing a COPD-on-chip model, stimulation conditions must be optimized to induce a reproducible exacerbation-like response without excessive cell damage. This study optimized viral stimulation conditions by comparing Poly(I), inactivated rhinovirus particles, and live rhinovirus in submerged Calu-3 monoculture, and Poly(I) in an ALI Calu-3/Jurkat co-culture. IL-6, IL-8, and IFN-β gene expression was measured at 6 and 24 hours using qPCR. Poly(I) induced the strongest and most reproducible response, mainly at 6 hours, while inactivated and live virus produced weaker and more variable responses. The ALI co-culture showed a different preliminary response pattern, although this was based on N=1. Overall, Poly(I) at 1–10 µg/mL appears most suitable for inducing a reproducible exacerbation-like response, while viral-particle and live-virus protocols require further optimization.
