Abstract

In pharmacokinetics, modeling the behavior of a drug is not always straightforward. There exist multiple possibly suitable parametrizations, but which is desirable/optimal? These questions are often difficult to answer. This thesis proposes more elaborate use of non-parametric methods. In particular, the generalized non-additive model (GAM) method is tested for three different data sets. The results are then compared to that of the traditional, parametric method of non-linear mixed effect modeling. Next to quality of fit, the quality of the models in terms of computational stability (and therefore also its reliability) is also considered for both types of models. The results show that the non-parametric method can be useful over parametric methods in certain cases and that further investigation on these circumstances and on the consequences on the interpretability trade-off of non-parametric methods have to be further examined.
