Abstract
Siderophores are small molecules who are used in the iron metabolism of various organisms like P. chrysogenum. The siderophores are made by Non-Ribosomal Protein Synthetases (NRPSs). Here we describe the effects of various iron-limited media upon the genes coding for the NRPSs involved in the biosynthesis of siderophores in P. chrysogenum. Iron-limited media was made by adding bathophenanthroline disulfonate (BPS) and by making are own media and omitting iron from the ingredients. P. chrysogenum was grown on iron containing and iron-limiting media and by transcript analysis the difference in activity was determined. Anti-smash analysis showed that the genes responsible for the iron metabolism pathway were clustered with their NRPS and transcript analysis showed that these genes respond together to iron-limiting conditions. The genes coding for NRPSs were showing a higher activity under iron-limited conditions than under standard conditions. Siderophores are needed to maintain iron homeostasis and when iron is limited in the environment the genes for NRPSs and their associated cluster respond to this limitation.
