Abstract
Tamoxifen is a hormonal antiestrogen currently used in the treatment of estrogen receptor-positive breast cancer. However, the potency of tamoxifen in tumor cells not expressing this receptor is low, and new strategies are needed for the treatment of these tumors. This report describes two possible strategies: light irradiation and tamoxifen co-treatment with histone deacetylase complex (HDAC) inhibitor SAHA. First, a two-step synthesis route to synthesize tamoxifen is described. Secondly, the effect of twenty minutes of light irradiation on the potency of tamoxifen was assessed in estrogen receptor-positive breast cancer cell line MCF-7, and light irradiation was not able to potentiate tamoxifen. However, at concentrations below 5.56 μM, a slight increase in cell proliferation was observed in response to light irradiation, indicating a mutagenic effect likely due to the formation of reactive intermediates. Lastly, the effect of co-treatment of tamoxifen and SAHA on the potency of SAHA was tested in MCF-7, Hel (leukemia), and SU-DHL-6 (lymphoma) cells. Moreover, tamoxifen and SAHA did not potentiate each other in these cell lines, demonstrating that tamoxifen and SAHA target different molecular pathways. Therefore, we failed in the development of a novel strategy to improve the potency of tamoxifen in estrogen receptor-positive breast cancer and liquid cancer.
